Dynamic Cross Linking (DXL) Technology, Antibodies with Increased Avidity
Our primary platform technology is Dynamic Cross Linking (DXL), which involves modification or design of antibodies with increased avidity and concomitant increases in complement dependent and cell dependent effector functions without altering affinity or specificity, thereby circumventing any new (unwanted or harmful) cross-reactions with unintended targets. Put simply: DXL results in safer antibodies with a higher level of therapeutic usefulness.

DXL antibodies have been shown, for example, to have increased ability to mediate non-effector killing of target tumor cells via apoptosis. For antibodies used in oncology, this means better killing of target tumors, and an ability to use less drug product versus an unmodified antibody. DXL antibodies have the potential to overcome resistance to certain widely used therapeutic antibodies by use of mechanisms not employed by the unmodified antibody. Studies of DXL625 (CD20) for the prospective treatment of non-Hodgkins Lymphoma for example, revealed no toxicity or immunogenicity in primates compared to a non-DXL control antibody, which confirmed results that were earlier indicated in smaller animal studies.
